ABSTRACT

Euphorbiae Ebracteolatae Radix is the dry root of plant Euphorbia fischeriana or E. ebracteolata of Euphorbiaceae, which is a widely utilized natural medicine with broad development prospect. It has been discovered that Euphorbiae Ebracteolatae Radix contains several biologically active constituents, among which diterpenoids are the most important. The diterpenoids of Euphorbiae Ebracteolatae Radix contain eight types including abietane-type, tigliane-type, pimarane-type, rosane-type, cembrane-type, ent-kaurane-type, ingenane-type and atisane-type. Diterpenoids of Euphorbiae Ebracteolatae Radix have significant anti-tumor, anti-inflammatory, anti-viral and other pharmacological activities. In this paper, the chemical constituents of diterpenoids and pharmacological activities of Euphorbiae Ebracteolatae Radix in recent years were reviewed in order to provide references for better developing the resources of Euphorbiae Ebracteolatae Radix and its clinical application.

ABSTRACT

To investigate the toxicity changes of Euphorbiae Ebracteolatae Radix (EER) before and after vinegar processing, toxic diterpenoids were concentrated with chloroform as extraction solvent from EER. Then the residue was extracted for non-chloroform extract with 95% ethanol and water after extraction with chloroform. The chloroform extraction of vinegar processed EER was prepared with the same method. The mice received the drug by oral administration. Moisture content in mice feces, duodenum and colon tissue, aquaporin AQP1, AQP3, AQP4 protein expression levels were assayed as the indexes to investigate the toxicity variation of chloroform fraction, non-chloroform fraction, as well as intestinal tract toxicity before and after vinegar processing of EER. The results showed that the chloroform fraction extracted from EER could significantly increase the moisture content in mice feces, duodenum and colon, and decrease AQP1 protein expression level, increase AQP3 and AQP4 protein expression levels in the colon. The intestinal toxicity of the chloroform extract was significantly higher than that of non-chloroform extract. The moisture content in mice feces, duodenum and colon was significantly decreased, and the AQPs protein expression tended to be normal in the colon after vinegar processing. The results showed that the chloroform fraction extracted from EER could lead to diarrhea, intestinal edema, and the intestinal toxicity action was associated with interfering AQPs protein expression and promoting intestinal fluid transport disorder in mice. Vinegar-processing could reduce intestinal toxicity of EER, so vinegar processing was considered to be the scientific processing method of EER.

ABSTRACT

In recent years, animal experiments and clinical trials have confirmed that Euphorbiae Ebracteolatae Radix has outstanding medicinal value of cancer prevention in the aspects of inhibition of tumor cell proliferation, inducing apoptosis, cell cycle regulation and regulation of immune. It also has been reported that monomeric compounds with function of anti-tumor activity have been separated. This article reviewed anti-cancer effects of Euphorbiae Ebracteolatae Radix from five angles, which were anti-cancer active ingredient, anti-cancer mechanisms, clinical application, the effects of different processed products on anti-cancer effects and side effects.

ABSTRACT

ObjectiveTo investigate effects ofEuphorbiae ebracteolatae Radixon the proliferation and apoptosis in human papillomavirus (HPV)-18-infected HeLa cells.MethodsThe alcohol extract ofEuphorbiae ebracteolatae Radixwas prepared. Cell proliferation was determined using the MTT assay, and apoptotic cells was determined using the Hoechst 33258 staining.ResultsHPV-18-infected HeLa cells were treated with 10, 20 μg/mlof the alcohol extract ofEuphorbiae ebracteolatae Radixfor 12 h, the cell proliferation inhibition rate were significantly increased compared to the control group (16.36% ± 5.06%, 18.59% ± 5.99%; allP<0.01);HPV-18-infected HeLa cells were treated with 0.625, 1.25, 2.5, 5, 10, 20 μg/ml of the alcohol extract ofEuphorbiae ebracteolatae Radixfor 72 h, the cell proliferation inhibition rate were significantly increased compared to the control group (13.82% ± 3.66%, 30.24% ± 6.58%, 43.29% ± 4.29%, 53.32% ± 1.41%, 54.24% ± 1.97%, 50.68% ±1.74%;allP<0.01). The Hoechst 33258 staining showed that HPV-18-infected HeLa cells were treated with 10 μg/ml of the alcohol extract ofEuphorbiae ebracteolatae Radixfor 24 h, the apoptotic cells were found.ConclusionEuphorbiae ebracteolatae Radixcould inhibit the proliferation and induce apoptosis in HPV-18-infected HeLa cells.
